There are two basic methods currently being used for the preparaticn of PPLO colonies for microscopic study. One methcd uses a modified agar fixation technique (Klieneberger and Smiles, J. Hyg., 42, 110, 1942) and the other a direct, stained agar technique (Dienes, J. Infectious Diseases, 65, 24, 1939). Both methods have been used and reported by other investigators to have
